This is done by determination of chlorophyll concentrations, transmission, and the photosynthetic activity of microalgae. These can be measured as total chlorophyll [µg chl-a/l], concentration of green algae [µg chl-a/l], concentration of cyanobacteria [µg chl-a/l], concentration of diatoma [µg chl-a/l] and concentration of cryptophytes [µg chl-a/l]’.

Algae sampling containers should be plastic bottles or plastic jars and should be half filled with liquid. Note that glass is reportedly not satisfactory for some Chrysophyta and other algae of acidic waters due to its inherent alkalinity damaging cells. For long-term storage, specimens can be preserved in liquid Commercial formalin (which is a solution of 40% formaldehyde), diluted between 1/10 and 1/20 with the collecting solution, is the most used fixative. Algae can be kept in diluted formalin for several years, but the solution is usually replaced by 70% ethyl alcohol with 5% glycerin (the latter to prevent accidental drying out).
